A jury of eleven will give the final verdict on the 2018 article written by Heard in The Washington Post.
In the article, she spoke about domestic violence towards women—implying Depp, her former husband, was an abuser.
The former couple has been caught up in a legal battle for six years since their divorce in 2016.
The hearing is in Fairfax County, Virginia, with witnesses from both sides.
British actor Paul Bettany will appear for Depp, who is expected to be present in court via video link. American actor James Franco and Tesla CEO Elon Musk are expected to appear for Heard.
